On April 26, 1982, Holmes was born in Campbellsville, Kentucky. He began his golf career while attending Taylor County High School. He competed on the Pepsi Junior Golf Tour throughout his high school career. Holmes subsequently transferred to the University of Kentucky, where he won the SEC Championship and represented the United States in the Walker Cup in 2005.
He became professional in 2005 and won the PGA Tour Qualifying Tournament that year. He went on to win the FBR Open the following year, becoming the first golfer on the PGA Tour to earn $1 million in his career. On his fourth PGA Tour, he was able to reach that level of success.
